What 1,260 Genetic Variants Reveal About Personality’s Reach Into Daily Life

Whether someone is the life of the party or the one who reads the room before speaking, science has long assumed personality comes down mostly to upbringing and life experience. A massive new genetic study of more than a million people suggests something else: the same genetic variants tied to traits like extraversion or openness also quietly shape a person’s health, career path, neighborhood and even how active they are late at night.
Published in Nature, the research pooled genetic and personality data from 46 research groups worldwide, focusing on the five major personality categories psychologists call the Big Five: extraversion, agreeableness, conscientiousness, neuroticism and openness to experience. Scientists scanned the DNA of somewhere between 611,037 and 1.14 million people per trait, hunting for spots in the genetic code tied to each dimension, then traced how those patterns connected to dozens of real-world outcomes.
“There is no single ‘personality gene,'” said Ted Schwaba, lead author of the study and a faculty member in Michigan State University’s Department of Psychology. Personality traits are tied to thousands of genetic variants, he said, and even the largest individual effects are tiny. Combined, those effects add up to something researchers could trace across a person’s life, from their doctor’s office to their dating history.
Scientists Found 1,260 Genetic Spots Tied to Personality
Researchers behind the project, called the Revived Genomics of Personality Consortium, pulled together data from many countries, including people with European and African genetic ancestry, plus a separate group of up to 50,725 participants used specifically to study genetics within families.
Combing through roughly 10 million genetic differences, scientists pinpointed 1,260 spots linked to one or more of the five traits, 824 of which, about 65%, had never before been linked to that particular trait. Neuroticism had the richest genetic map by far, with 706 associated spots, compared to just 39 for agreeableness.
Genetic effects held up remarkably well across very different people. Researchers found that a genetic variant tied to extraversion in a young adult in the Netherlands showed the same link in an older retiree in the United States. Effects were similarly consistent whether personality was self-reported or described by a close friend, and across the US, continental Europe, the Nordic countries and the combined UK and Australia. Common genetic differences still explained only a modest slice of the personality puzzle, between 4.8% and 9.3% of the differences between people in each trait, and environment still plays a major role in shaping who someone becomes.
Personality Genes Predict Health Risks and Hospital Visits
Genetic variants tied to neuroticism carried higher genetic risk across every form of mental health condition studied, especially anxiety and depression-like low mood. Schwaba said those same variants predict more time spent in the hospital. Agreeableness ran the opposite direction, linked to lower risk across many of those same conditions.
Conscientiousness genetics lined up with lower substance use, more exercise, a preference for lower-calorie foods and healthier weight. Variants linked to conscientiousness also predicted being more active during the day, while variants tied to openness to experience were linked to being more active at night, a pattern picked up using wearable activity monitors.
Openness Genes Point Toward City Life and Job Switching
One of the study’s more striking threads involves where people end up living and working. Genetic variants linked to openness predicted a greater likelihood of moving into cosmopolitan city neighborhoods later in life, while variants linked to conscientiousness predicted the opposite, a move toward affluent suburban communities.
Openness-linked genetics were also tied to higher income and a greater likelihood of switching jobs. Schwaba said the pattern likely runs in both directions. A genetic pull toward openness might lead someone into a lively city neighborhood, and living there could then reinforce and further shape the same trait, an interconnected loop between genes and environment rather than a one-way street.
The Genetic Link Isn’t Explained by Family or Romantic Pairing
Researchers also tested a long-standing assumption: that people pair off with romantic partners who share their genetic tendencies toward certain personality traits. Using genetic data on spouses, siblings and cousins from the UK Biobank and Iceland’s deCODE database, they found genetic similarity between spouses was minimal, just 1% to 4%. That similarity was far stronger for educational attainment, around 37%, than for personality. A separate check on the personality-DNA link itself, using a genetic database in Iceland covering more than 34,000 parent-child pairs, found direct genetic effects accounted for an average of 96.2% of the total predicted effect, confirming the link was not primarily a byproduct of growing up alongside a parent who happened to share similar genes.
None of this means a DNA test can predict what any one person will do with their life. These are broad trends visible only across huge numbers of people, not a way to read someone’s future from a cheek swab.
Schwaba said he hopes the findings push researchers well outside genetics to take personality more seriously. Neuroticism’s link to hospital visits matters to physicians, he said, while people who study careers might take note of openness’s ties to the labor market. Genes may set the stage, but where someone lives and what they do for work both seem to keep shaping the trait right back.

Limitations
Discovery work relied heavily on European-like genetic ancestry, with fewer African-like cohorts, meaning findings may generalize less well to other ancestral groups. Within-family analyses, while showing minimal confounding, used smaller sample sizes than population-level analyses, widening uncertainty in some estimates. Because personality is influenced by so many genetic variants at once, any single variant’s effect on its own was too small to be meaningful, and genetic prediction scores explained only a small share of variance in personality and related outcomes, so they cannot predict an individual’s future behavior. Agreeableness showed less consistent genetic signal across cohorts, instruments and raters than the other four traits, warranting extra caution in interpreting results for that trait.
